Not in position one, and not more than one in the set. A photo of you with someone of the gender you are looking for creates an unanswerable question - who is that - and a stranger will not spend a swipe resolving it.
Further down the set, one such photo is fine and can even help, because it shows you have friendships rather than only portraits.

The opening card is one photo and a name. Two people in it means the viewer has to work out which one you are before they can decide anything, and that is more effort than the moment allows.
Worse, if the other person is more striking, that is who the card is now about. This is the same failure as a group shot in position one, and it is expensive for the same reason.
The common reads are: that is an ex, that is a partner, or this is a photo chosen to signal being wanted. None of them is necessarily true and all of them are cheap to trigger.
You cannot caption your way out of it on most apps, and a bio line explaining who is in photo three is a bad use of your two good lines.
Position four or five, one of them, in a photo that is obviously social rather than a couple shot - a group of friends, an event, something happening. Then it reads as a life, which is what a social photo is for.
Crop out an ex rather than keeping the photo because you like how you look in it. That is the most common reason these photos survive on a profile, and it is not a good enough one.
A photo with a sibling or a parent is usually read correctly and rarely causes a problem, because family resemblance does the explaining.
